New England Biolabs, Inc. PaqCI - 1000 units
PaqCI is a Type IIS restriction enzyme with a 7 basepair recognition sequence: CACCTGC(4/8). It is sourced from an E. coli strain that carries the modified cloned PaqCI gene from Paucibacter aquatile. PaqCI requires multiple sites for efficient cleavage -- PaqCI Activator is included for optimal cleavage. This enzyme is supplied with NEB's rCutSmart Buffer and is heat inactivated.
New England Biolabs, Inc. EnGen Spy Cas9 Nickase – 70 pmol
EnGen Spy Cas9 Nickase is a variant of Cas9 nuclease differing by a point mutation (D10A) in the RuvC nuclease domain, which enables it to nick, but not cleave, DNA). EnGen Spy Cas9 Nickase, like EnGen Cas9 NLS , targets DNA using a guide RNA complementary to a site with a 3' NGG protospacer adjacent motif (PAM). Nicking occurs on the DNA strand opposite the PAM. Double-stranded DNA breaks can be generated with reduced off-target cleavage by targeting two sites in close proximity (generally 0-20 bp apart) and with PAMs facing outward to leave 5' overhangs. Nicking by EnGen Spy Cas9 Nickase occurs on the DNA strand opposite the target sequence. EnGen Spy Cas9 Nickase contains SV40 T antigen nuclear localization (NLS) sequences on the N and C- termini.

New England Biolabs, Inc. RNase Inhibitor, Human Placenta – 2000 units
RNase Inhibitor, Human Placenta is a recombinant human placental protein which specifically inhibits ribonucleases (RNases) A, B and C. It is not effective against RNase 1, RNase T1, S1 Nuclease, RNase H or RNase from Aspergillus. In addition, no inhibition of polymerase activity is observed when RNase Inhibitor, Human Placenta is used with Taq DNA Polymerase, AMV or M-MuLV Reverse Transcriptases, or Phage RNA Polymerases (SP6, T7, or T3). The 50 kDa protein inhibits RNases by binding noncovalently in a 1:1 ratio with an association constant greater than 1014.

A high fidelity restriction endonuclease that recognizes the sequence CG_AT^CG. High Fidelity (HF) Restriction Enzymes have 100% activity in rCutSmart Buffer; single-buffer simplicity means more straightforward and streamlined sample processing. HF enzymes also exhibit dramatically reduced star activity. HF enzymes are all Time-Saver qualified and can therefore cut substrate DNA in 5-15 with the flexibility to digest overnight without degradation to DNA. Engineered with performance in mind, HF restriction enzymes are fully active under a broader range of conditions, minimizing off-target products, while offering flexibility in experimental design.
